Description
The ExcelTaq™ 2X Fast Q-PCR Master Mix (SYBR, ROX) is a ready-to-use reagent with all the essential components for quantitative real-time PCR (qPCR) except primers and templates. The master mix contains a highly stable hot-start Taq polymerase in an optimized buffer with dsDNA specific SYBR green fluorescent dye. Consequently, the ExcelTaq™ 2X Fast Q-PCR Master Mix (SYBR, ROX) features high stability during storage, even at 37°C for weeks. In addition to high sensitivity and signal intensity, the ExcelTaq™ 2X Fast Q-PCR Master Mix (SYBR, ROX) performs a low background/ high specificity qPCR results, as well as a better compatibility with fast PCR program. With inert smart blue contrast dye, the ExcelTaq™ 2X Fast Q-PCR Master Mix (SYBR, ROX) is ready-to-use and greatly reduces pipetting errors, while largely improving the reproducibility of the process. The ExcelTaq™ 2X Fast Q-PCR Master Mix (SYBR, ROX) also includes ROX reference dye for normalizing the fluorescent reporter signal in real-time quantitative PCR. This master mix allows sensitive, precise amplification, real-time tracking of the amplification process, and simultaneous quantification for targeted DNA molecules.

Fast Hot start and better efficiency
The standard curves were examined using fast program with cDNA ranging from 200 fg to 20 ng as template. The efficiency was calculated according to the slope of standard curve. The efficiency of TQ1210 is approximately 100% showing that TQ1210 is more compatible with fast program as compared to brand A (efficiency: 71.9).
High Sensitivity
The sensitivity assay of TQ1210 ExcelTaq™ 2X Fast Q-PCR Master Mix (SYBR, ROX) was examined with serially diluted cDNA from 25 ng to 1.4 fg as templates. The amplification curves display that TQ1210 is capable to amplify template as little as 1.4 fg, and over a wide range of template usage.
Low nonspecific product
The post-amplification melt-curve analyzed with relatively higher concentration of primers (400 nM) reveals the specificity of qPCR reagents. The multiple peaks of melt curve display a nonspecific and a target PCR products. The ratio of nonspecific PCR product to target PCR product represents the specificity of qPCR reagents. The ratio analyzed using TQ1210 (0.5 to 1) was lower than the ratio analyzed from brand P (2 to 1) and A (0.8 to 1), showing high specificity of TQ1210.
High Stability
Stability was analyzed via real-time PCR amplification with 2 ng, 20 pg and 0.2 pg cDNA as template, and TQ1210 ExcelTaq™ 2X Fast Q-PCR Master Mix (SYBR, ROX) stored at -20°C or 37°C for 4 weeks as reagents. The overlapped amplification curves of qPCR reactions from different storage conditions display excellent stability of TQ1210 ExcelTaq™ 2X Fast Q-PCR Master Mix (SYBR, ROX). Inconsistent Ct value and decreased fluorescent intensity were observed in qPCR master mix from brand P.

TQ1210 can be compatible with fast program and save 60% of run time (<1hr).
